The Blues must come from a 3-1 deficit to secure their place in the semi-finals of the competition after a Karim Benzema hat-trick gave Carlo Ancelotti’s side a huge lead last week.
Chelsea recovered from their European Cup defeat and defeated Southampton 6-0 at St Mary’s at the weekend.
But neither Lukaku nor Azpilicueta were included in the Premier League matchday squad against the Saints.
Striker Lukaku is still suffering from Achilles tendon pain and will not make the trip to the Bernabeu.
Club captain Azpilicueta, who tested positive for Covid ahead of the Southampton clash, has now tested negative multiple times and is in the squad against Madrid.
‘Callum Hudson-Odoi is still out, Ben Chilwell is still out, Romelu Lukaku will not travel with us because of his Achilles tendon pain,” Tuchel told reporters on Monday.
‘Azpilicueta has tested negative multiple times and is in the group. Ross Barkley is ill and will not be traveling.’
Regarding his team’s chances of turning the tie around, Tuchel said: “Not the greatest chance given the results of the first game and given the competition, the opponent and the stadium.
“We never manage our input based on the opportunities we have for our outcome.
“It’s unlikely but it’s worth a try and trying means we’re going to play our best and our full limit. That’s it, it’s a big night.’
