Antony, 22, has been one of Erik ten Hag’s top targets all summer after starring under the Dutchman in two joint title-winning seasons in Amsterdam.
However, Ajax were keen to keep the Brazil international and raised their asking price for him earlier in the summer, which seemed to blunt interest from United.
Athletic report United returned on Wednesday with a huge offer to test Ajax’s resolve.
However, another rejection is likely to see the Premier League side end their interest in a deal this summer, as the report adds club officials are “not optimistic” about finalizing a deal.
United see PSV Eindhoven star Cody Gakpo as an alternative option and see his signing as far more likely of the two.
The Premier League side are preparing a £35m bid for the versatile forward, with reports in the Netherlands last week that ten Hag have already been in touch about a move.
However, Gakpo’s future could depend on PSV’s promotion to the Champions League. The Dutch giants sealed a 4-3 aggregate win over Monaco in their third qualifying round tie and drew in this week’s play-off first leg against Rangers.
A win in the second leg next week secures PSV a place in the group stage of the competition.
Gakpo, who plays mostly on the left wing at PSV but can also act in the middle, is represented by Dutch agency SEG, which also advises Ten Hag.
United are also considering a loan move for Chelsea striker Christian Pulisic, with the American interested in a move to Old Trafford to secure greater first-team opportunities.
However, the Blues are reportedly unwilling to authorize a move to a Premier League rival.
Atletico Madrid’s Yannick Carrasco is another option the club are considering.
